Overview

FYVE and coiled-coil domain autophagy adaptor 1 (FYCO1) is a cytosolic Rab7 adaptor protein that mediates microtubule plus end-directed transport of autophagic vesicles by interacting with several key partners, including the small GTPase Rab7, phosphatidylinositol-3-phosphate (PI3P), the autophagosome marker LC3, and kinesin KIF5. It contains a RUN domain, FYVE-type zinc finger domain, and GOLD domain. FYCO1 is essential for proper autophagosome movement and maturation, playing a central role in autophagy. Genetic mutations in FYCO1 cause autosomal recessive congenital cataracts and are associated with rare forms of muscle disease such as inclusion body myositis. Its dysfunction may also alter cell migration and invasion processes, hinting at a broader biological relevance.
