Overview

G antigen 1 (GAGE1) is a member of the cancer/testis antigen family, genes typically expressed in various malignancies and in normal testis but not in most other normal tissues[6][4]. The protein is recognized as an antigen by autologous cytolytic T lymphocytes in certain cancers, notably melanoma, and has been investigated as a target for cancer immunotherapy due to its restricted expression pattern and immunogenicity[1][6]. The biological function of GAGE1 remains largely unknown, though it is implicated in cellular defense responses, possible resistance to apoptosis, and cancer cell survival[4][5]. While frequently upregulated in cancer and correlated with tumor progression, existing data do not definitively establish a role for GAGE1 in metastasis[2][5]. Expression can be induced by epigenetic modification in some tumor models. GAGE1’s value lies primarily as a tumor-associated antigen and a potential target for T cell–based immune therapy in cancers expressing this protein, but therapeutic strategies are limited by variable expression and incomplete functional characterization[1][4][5][6].
