G antigen 12B (GAGE12B) is a protein-coding gene and member of the GAGE family of cancer/testis antigens. These proteins are notable for being expressed almost exclusively in human testis and a wide variety of tumor types, while showing very limited or no expression in other normal tissues. Like other GAGE family proteins, GAGE12B is relatively small, predicted to be composed of approximately 117 amino acids. The family is characterized by sequence similarity, clustered location on chromosome Xp11.23, and poorly understood structure and biological functions, classifying them as part of the cellular "dark proteome" where much remains to be elucidated regarding their precise molecular interactions and physiological roles. GAGE antigens, including GAGE12B, are of interest as potential immunotherapeutic targets and biomarkers for cancer, but no direct drug interactions or clinical development targeting GAGE12B have been established to date.
