Overview

G antigen 2A (GAGE2A) is a protein encoded by the GAGE2A gene located on the human X chromosome. It is a cancer/testis antigen, meaning its expression is largely restricted to germ cells and many types of human cancers, but virtually absent in normal somatic tissues. These antigens are immunogenic and can elicit cytotoxic T-cell responses. GAGE2A, along with other GAGE family members, has been investigated as a potential biomarker for cancer prognosis and as a target for immunotherapy, especially cancer vaccines. Elevated mRNA levels of GAGE2A have been associated with poor prognosis in ovarian cancer and potentially other malignancies. The gene sequence is highly conserved among family members, differing only by scattered nucleotide changes, and encodes peptide antigens recognized by cytotoxic T cells. There are currently no approved drugs directly targeting GAGE2A, but it remains a significant candidate for biomarker-based patient selection and targeted immunotherapy research.

For hypothetical immunotherapeutics: immune recognition and cytolytic T-cell targeting of tumor cells expressing GAGE2A peptide antigens. Mechanism for drugs would involve induction of immune-mediated tumor cell killing.
