Overview

G patch domain-containing protein 1 (GPATCH1) is a highly conserved RNA-binding protein distinguished by its N-terminal G-patch domain, which interacts with spliceosomal proteins and RNA[1][3]. GPATCH1 serves as a regulatory cofactor for the DEAH-box RNA helicase DHX35, anchoring it to the active spliceosome and facilitating proper pre-mRNA splicing quality control by suppressing aberrant splicing events[1]. Structural and functional studies demonstrate that GPATCH1 interacts with core spliceosome components (notably PRP8), recognizes suboptimal splice sites, and helps maintain splicing fidelity across eukaryotes from yeast to humans[1][2]. There are currently no known drugs or targeted therapies acting directly on GPATCH1, nor is it established as a classical therapeutic target or disease biomarker.
