Overview

G-patch domain-containing protein 3 (GPATCH3) is a member of the G-patch protein family characterized by a glycine-rich G-patch motif that enables interaction with DEAH-box RNA helicases, especially DHX15[3]. It acts primarily as a modulator of RNA splicing, facilitating spliceosome disassembly and proper alternative splicing through enhancing DHX15 ATPase activity[1][3]. GPATCH3 is implicated in regulating immune processes within the tumor microenvironment, promoting immunosuppressive cell populations and facilitating tumor immune evasion when overexpressed[1][3]. It has also been shown to negatively regulate RIG-I-like receptor-mediated antiviral innate immune signaling, acting on the mitochondrial antiviral signaling protein VISA/MAVS[4]. High GPATCH3 expression correlates with poor prognosis in several cancers, and loss of GPATCH3 in preclinical models enhances immune cell infiltration and suppresses tumor growth[1][3]. In addition, GPATCH3 is expressed in ocular tissues and rare genetic variants have been observed in congenital glaucoma patients, suggesting possible relevance for ocular development and function[6]. There are no known drugs that directly interact with GPATCH3, and it is not currently considered a biomarker or therapeutic drug target in clinical use.
