Overview

G protein-coupled receptor 137 (GPR137) is an integral membrane protein that functions as a cell surface receptor. It belongs to the GPCR superfamily and mediates signal transduction processes involved in cell growth, cell cycle regulation, and apoptosis. GPR137 is expressed in various tissues, notably in the brain, kidney, liver, and is highly expressed in certain cancer cell lines. Recent studies show that silencing GPR137 inhibits cell proliferation and triggers apoptosis in leukemia and gastric cancer cells, identifying it as a promising therapeutic target and biomarker for cancer diagnosis and therapy. GPR137 has three splice isoforms (GPR137A, GPR137B, and GPR137C), of which GPR137A is most often referenced as GPR137. Its precise endogenous ligand and broader physiological role remain to be elucidated; however, its oncogenic potential is increasingly recognized.
