Overview

G protein-coupled receptor 155 (GPR155) is an integral membrane protein encoded by the GPR155 gene in humans[4]. It is an atypical GPCR featuring a unique 17-transmembrane domain architecture, combining a transporter-like domain (10 TM) and a classical 7TM GPCR region, along with C-terminal DEP domains[2]. GPR155 binds cholesterol in lysosomal membranes, acting as a sensor to regulate mTORC1 signaling—promoting growth signaling when cholesterol is abundant[3][5]. In cancer (notably gastric cancer), reduced GPR155 expression is linked to increased metastasis and cell invasiveness; its mRNA level serves as a predictive biomarker for hematogenous spread[1][6]. Mutations or altered expression may be associated with autism spectrum disorder and other neurodevelopmental conditions[4]. There is no credible evidence for a distinct “divergent transcript” or for drug interactions with GPR155 to date.
