G protein-coupled receptor 182 (GPR182) is a highly conserved membrane protein primarily expressed in the vascular and lymphatic endothelium. Although originally identified as a putative adrenomedullin receptor, it is now recognized as an atypical chemokine receptor that functions as a scavenger for ligands such as CXCL12, CXCL11, and CXCL13. By internalizing these chemokines, GPR182 regulates their extracellular concentrations and gradients, thereby modulating processes like angiogenesis, leukocyte trafficking, and hematopoietic stem cell maintenance. In the context of disease, GPR182 is often downregulated in hypervascular tumors like hepatocellular carcinoma, where its loss leads to excessive CXCR4-mediated vessel sprouting and tumor progression. Consequently, it is being investigated as a therapeutic target for anti-angiogenic strategies and as a regulator of the immune microenvironment in cancer.
Acts as a decoy or scavenger receptor that internalizes and degrades chemokines (CXCL12, CXCL11, CXCL13) to regulate their extracellular gradients and limit signaling through their primary receptors (e.g., CXCR4).
