GPR183 (EBI2) is a chemotactic G protein-coupled receptor that plays a central role in the positioning, migration, and interaction of immune cells. It is highly expressed in immune system tissues and cells, including B cells, T cells, dendritic cells, monocytes, macrophages, and astrocytes. Activation of GPR183 by oxysterols, particularly 7α,25-dihydroxycholesterol (7α,25-OHC), guides immune cell movement within lymphoid tissues and contributes to effective adaptive immunity. Dysregulation of GPR183 signaling is implicated in viral infection-related inflammation and leukemia.
Ligand binding initiates Gi-alpha signaling, leading to downstream effects including MAPK/ERK pathway activation. Chemotaxis is a primary downstream effect.
