G protein-coupled receptor 25 (GPR25) is an orphan receptor belonging to the class A (rhodopsin-like) family of G protein-coupled receptors[1][2][3][6][8]. GPR25 is encoded by an intronless gene and is highly conserved evolutionarily[1][2]. In humans, its expression is enriched in lymphocytes, natural killer (NK) cells, some T-cells, and to a lesser degree in the stomach[1][7]. Its biological functions are incompletely characterized, but evidence implicates GPR25 in signal transduction pathways affecting immune response and blood pressure regulation, with genetic association studies linking GPR25 to arterial stiffness and psoriasis[2][3][7]. GPR25 is classified as an orphan receptor because a definitive endogenous ligand in humans is not confirmed, though it can be activated by certain peptides in non-mammalian vertebrates and may have chemokine receptor-like properties[2][1]. No drugs are currently known to target GPR25, and there are no established biomarkers or safety concerns at this time.
