The 12-hydroxyeicosatetraenoic acid receptor is a high-affinity, G protein-coupled receptor (GPCR) that specifically binds the eicosanoid 12-(S)-hydroxy-5,8,10,14-eicosatetraenoic acid (12-S-HETE). Activation of this receptor leads to activation of several intracellular signaling pathways, promoting cell growth, proliferation, survival, and macropinocytosis. It has been implicated in cancer, cardiovascular disease, renal disease, and immune system modulation.
Upon binding of 12-(S)-HETE, the receptor activates downstream signaling pathways including ERK1/2, MEK, and NF-kappa-B, leading to changes in cell growth, proliferation, survival and other cellular processes. A cytosolic/nuclear complex may also be involved.
