Overview

GPR35 is an orphan GPCR with high expression in immune/gastrointestinal tissues. It modulates inflammation through complex mechanisms that can be either pro-or anti-inflammatory based on cellular context. Its involvement extends into cardiovascular regulation and cancer biology—making it a promising therapeutic target despite challenges related to ligand specificity/species selectivity among tool compounds currently available for research use.

Mechanism of action

GPR35 activation induces conformational change leading to activation of associated heterotrimeric G proteins. Downstream signaling influences immune cell activation/chemotaxis/cytokine production or suppression depending on cellular context. Modulates angiogenesis within tumor microenvironments.
