G protein-coupled receptor associated sorting protein family member 3 (GPRASP3) is a basic helix-loop-helix domain-containing protein involved in multiple aspects of neuronal development, survival, and differentiation. It acts by modulating neurosynaptogenesis, inducing phosphatase PP2A activity (promoting APP dephosphorylation and inhibiting amyloidogenic BACE1 processing of APP), and, as a GPCR-associated sorting protein, may control the trafficking and signaling of G protein-coupled receptors. GPRASP3 interacts with cellular signaling pathways critical to both neurodegeneration (notably Alzheimer's disease) and oncogenesis (with distinct mutation hotspots influencing cancer risk). Its activity is important for maintaining the survival/apoptosis balance in neural tissues and may be involved in aging processes. Multiple alternatively spliced transcript variants encode the same protein, and its expression is subject to regulatory perturbations by various drugs and chemicals according to gene expression datasets.
