Overview

G protein-coupled receptor class 5 member D (GPRC5D) is a class C orphan GPCR predominantly expressed in multiple myeloma cells and hard keratinized tissues like hair and nails. Its limited expression in most normal human tissues makes it an attractive therapeutic target for multiple myeloma. Several immunotherapies, including bispecific antibodies and CAR T-cell therapies, are being developed against GPRC5D. Overexpression in bone marrow correlates with poor prognosis for MM patients.

Mechanism of action

Redirects T cells to kill GPRC5D+ MM cells by binding both CD3 on T cells & GPRC5D; Binds ECLs/TM domains of dimeric GPRC5D; Engineered T cells targeting surface-expressed GPRC5D
