Overview

GPRC5D is an orphan G protein-coupled receptor (GPCR) belonging to the class C family. It is predominantly expressed in hard keratinized tissues like hair and nails, and highly expressed in multiple myeloma cells, but shows low expression in most other normal human tissues. While its exact biological function remains unclear, it is implicated in hair and nail development. GPRC5D is a therapeutic target for multiple myeloma, with immunotherapies like CAR T-cell therapy and bispecific antibodies (e.g., talquetamab) showing promising efficacy due to its restricted expression profile, which reduces off-target effects. Structural studies reveal GPRC5D forms dimers, influencing drug binding characteristics.
