Overview

The GRK2–delta-opioid receptor protein-protein interface is the molecular region where G protein-coupled receptor kinase 2 (GRK2) interacts physically with the delta-opioid receptor (DOR, a GPCR). GRK2 is a serine/threonine kinase involved in the phosphorylation and desensitization of activated GPCRs, including opioid receptors. Upon interaction, GRK2 phosphorylates the activated DOR, initiating receptor desensitization and internalization, which modulates opioid signaling and analgesic response. The precise structure and role of the interface are subjects of research as part of efforts to understand and manipulate opioid signal regulation, pain control, and related disease pathways. Currently, the interface itself is a subject of mechanistic study rather than a direct drug target.

Mechanism of action

GRK2 inhibitors block the phosphorylation/desensitization of GPCRs, potentially enhancing receptor sensitivity. DOR agonists or antagonists modulate opioid receptor activity. Inhibition of protein–protein interaction could theoretically modulate opioid signaling, but no approved drugs do so.
