Overview

G-protein-signaling modulator 2 (GPSM2) is a crucial regulatory protein that modulates heterotrimeric G protein activation and is essential for correct orientation of the mitotic spindle and maintenance of cell polarity during cell division[1][3][4]. It contains 10 Leu-Gly-Asn repeats in its N-terminal domain and 4 GoLoco motifs in the C-terminal, which confer guanine nucleotide dissociation inhibitory activity[1][4]. GPSM2 plays pivotal roles in multiple cellular processes, including neuroblast division and inner ear development. Its expression and post-translational modification (phosphorylation) have been associated with tumorigenesis, progression, and prognostic outcome in several cancers, notably liver hepatocellular carcinoma and breast cancer. Mutations are causative for some inherited disorders, such as nonsyndromic sensorineural deafness (DFNB82) and Chudley-McCullough syndrome[4]. Emerging evidence proposes GPSM2 as a molecular target under experimental investigation for cancer therapy, though no clinically approved drugs currently exploit this mechanism[3][4].
