G protein signaling modulator 3 (GPSM3) is a cytoplasmic protein highly expressed in leukocytes that regulates heterotrimeric G protein signaling downstream of G protein-coupled receptors (GPCRs). It contains GoLoco motifs enabling it to interact with Gα_i and Gβ subunits, functioning as a guanine nucleotide dissociation inhibitor (GDI) and thus modulating GPCR signal strength and duration. GPSM3 plays a critical role in immune cell migration and inflammation and is implicated as both a biomarker and a putative therapeutic target for autoimmune and inflammatory diseases, especially rheumatoid arthritis[1][2][3][4][5][7]. Genome-wide association studies have linked GPSM3 polymorphisms to autoimmune conditions, further supporting its relevance in disease biology[1][3]. No known direct small molecule or biologic drug interactions have yet been reported for GPSM3 as of current knowledge[2][3].
