Overview

The heterotrimeric G protein beta-gamma complex, particularly the Gβ1γ2 isoform, is a critical signaling component that dissociates from the Gα subunit upon activation of G protein-coupled receptors (GPCRs). While historically viewed as a negative regulator of Gα, the Gβγ dimer is now recognized as a potent signaling molecule that directly modulates various effectors, including ion channels (e.g., GIRK, Cav2.2), enzymes (e.g., PLCβ, PI3K), and kinases (e.g., MAPK). Dysregulation of Gβγ signaling is implicated in diverse pathologies, including cancer metastasis, heart failure, and chronic inflammation. Furthermore, germline mutations in the GNB1 gene (encoding Gβ1) lead to a severe neurodevelopmental disorder known as GNB1 encephalopathy. Therapeutic targeting of Gβγ focuses on small molecules like Gallein and M119, which disrupt the "hot spot" interface where Gβγ interacts with its effectors, offering a strategy to selectively inhibit downstream signaling without completely abolishing GPCR function. However, the ubiquitous nature of Gβγ and its central role in the G protein cycle present significant challenges for drug development, particularly regarding systemic toxicity and off-target effects.
