G protein subunit beta 5 (GNB5) is a regulatory protein that forms part of the heterotrimeric guanine nucleotide-binding (G) protein complex, functioning primarily to mediate and regulate signal transduction initiated by G protein-coupled receptors (GPCRs). It modulates signaling by regulating the GTPase activities of regulators of G protein signaling (RGS) proteins, especially RGS7 and RGS9, which are critical for the termination of G protein signaling in the nervous system, mood and cognition, and heart rate control. Mutations in GNB5 are associated with neurodevelopmental syndromes presenting intellectual disability, developmental delay, and cardiac arrhythmias.
