G-quadruplexes (G4s) are non-canonical, four-stranded secondary structures formed by guanine-rich sequences in nucleic acids. They are built around stacks of guanine tetrads, stabilized by monovalent cations, and exhibit diverse topologies. G4s are involved in key genomic processes and are considered promising molecular targets for drug discovery, particularly in cancer therapy. Several small-molecule ligands have been developed to selectively bind/stabilize G4s.
