Overview

GA-binding protein transcription factor subunit beta-1 antisense RNA 1 (GABPB1-AS1) is a cytoplasmic long non-coding RNA located at the GABPB1 gene locus on chromosome 15q21.2 that forms RNA duplexes with GABPB1 mRNA, modulating its translation by blocking polysome recruitment and regulating its interaction with the eukaryotic initiation factor eIF4A. GABPB1-AS1 also acts as a competitive endogenous RNA (ceRNA), sponging microRNAs such as miR-330 and miR-641, thereby influencing the expression of downstream targets such as ZNF367 and NUCKS1, which affect cell cycle, apoptosis, and cancer cell behavior. Upregulation of GABPB1-AS1 is associated with tumor malignancy, poor prognosis in glioma, and altered stress responses in ischemic injury models. Knockdown approaches targeting GABPB1-AS1 promote cell apoptosis, reduce cancer cell proliferation, and mitigate cerebral ischemic damage, identifying it as a valid molecular target for therapeutic intervention.

Mechanism of action

Inhibition of GABPB1-AS1 expression via RNA interference leads to upregulation of GABPB1 protein and downstream antioxidant proteins, decreased malignant cell proliferation, increased apoptosis, and protection against ischemic damage. Sponging of specific microRNAs (e.g., miR-330, miR-641) via GABPB1-AS1 alters gene regulatory networks involved in cell cycle and apoptosis.
