Overview

The HIV-1 Gag protein is a polyprotein precursor essential for the assembly, budding, and release of new virions from infected cells. It is both necessary and sufficient to drive the formation of virus-like particles, even in the absence of other viral proteins. The Gag polyprotein orchestrates several critical steps in the HIV-1 life cycle: directs virion assembly at the plasma membrane, mediates incorporation of viral RNA genome and other components into nascent particles and undergoes proteolytic cleavage to yield mature structural proteins required for infectious virus production. The full-length HIV-1 Gag polyprotein (Pr55^Gag) consists of several domains arranged linearly: MA (Matrix), CA (Capsid), NC (Nucleocapsid), SP1, SP2 and p6. During or shortly after budding, the viral protease cleaves Gag at five specific sites to generate these mature proteins.
