Overview

Galectin-3-binding protein (LGALS3BP), also known as the 90K tumor-associated antigen or OC183B2 antigen, is a large, multifunctional secreted glycoprotein that is significantly overexpressed in various human malignancies, including ovarian, breast, and lung carcinomas (Mattes et al., 1991). It was originally identified as the target of the monoclonal antibody OC183B2, which recognizes a specific epitope on this high-molecular-weight protein (Iacobelli et al., 1993). Biologically, LGALS3BP plays a critical role in the tumor microenvironment by mediating cell-cell and cell-matrix interactions through its binding to galectins, integrins, and fibronectin, thereby promoting tumor cell adhesion, migration, and metastasis (UniProt P17931). Additionally, it has been implicated in modulating the host immune response and promoting pro-survival signaling pathways in cancer cells (Grassadonia et al., 2004). Because of its high expression in malignant tissues and its presence in the serum of cancer patients, LGALS3BP serves as both a valuable prognostic biomarker and a potential therapeutic target. Experimental strategies have utilized antibodies like OC183B2 to develop immunotoxins, such as OC183B2-PE40, aimed at selectively eliminating cancer cells expressing this antigen. The protein's ability to form large oligomeric complexes further enhances its role as a molecular scaffold in the extracellular space. Clinical interest in LGALS3BP continues to grow as its role in viral infections and chronic inflammation is further elucidated. Despite its potential, therapeutic development faces challenges related to the protein's presence in normal tissues and its complex biological roles. Overall, LGALS3BP represents a significant target for both diagnostic and therapeutic interventions in oncology.
