Gametocyte development protein 3 (MD3)

Target
MD3
Molecular classification
RNA-binding protein, CCCH-type zinc finger protein, Post-transcriptional regulator
01

Overview

Gametocyte development protein 3 (MD3) is a cytoplasmic CCCH zinc finger protein found in immature male gametocytes of Plasmodium falciparum. It plays a crucial regulatory role in male gametocytogenesis by interacting with an array of other RNA-binding proteins, including translational regulators (eIF4G, PUF1, NOT1, CITH) and further regulators of gametocyte differentiation (ZNF4, MD1, GD1). MD3 is necessary for proper gametocyte maturation, and parasites lacking MD3 exhibit substantial impairment in the development of male gametocytes.

Mechanism of action

Not directly targeted by drugs yet; inactivation or deficiency leads to impaired gametocyte (specifically male gametocyte) maturation and thus blocks transmission of malaria from human to mosquito

Biological functions

Regulation of male gametocytogenesisPost-transcriptional gene regulation in gametocyte maturationInteraction with other RNA-binding proteins involved in translational regulation

05

Safety considerations

Not established; as a target in malaria parasites and absent in humans, targeting is theoretically highly specific but practical drug development data are lacking
