Overview

Gametocyte-specific factor 1-like (GTSF1L) is a paralog of GTSF1, expressed specifically in gonocytes and likely conserved in various species, particularly insects and mammals. GTSF1L features two CHHC-type zinc finger domains at its N-terminus, which facilitate RNA binding. Unlike GTSF1, GTSF1L associates with PIWI proteins such as MIWI and MILI via its N-terminal domain, and this interaction is RNA-dependent[3]. In silkworms and other insects, GTSF1L has evolved to orthogonally activate target RNA cleavage by specific PIWI proteins, promoting the production of piRNAs in vivo[1]. This specialization appears to be conserved among Lepidoptera and certain mosquitos. GTSF1L does not itself function as a known therapeutic target, nor are there drugs reported to interact with it. Experimental evidence shows its role is critical in stabilizing PIWI protein conformation for effective RNA slicing and facilitating protein-protein interactions important in germ cell biology[1][3][4]. To date, no direct association with human disease or utility in clinical diagnostics has been established.
