Overview

Gamma-adducin is a membrane-cytoskeleton-associated protein, part of the adducin family, that promotes assembly and stabilization of the spectrin-actin network in cells, essential for cell membrane integrity, cell-cell contact, and mechanosensation. It is expressed ubiquitously except for beta-adducin, and plays critical roles in vascular smooth muscle cell function, podocyte structure, blood pressure regulation, cell adhesion, and migration. Alternative splicing generates multiple isoforms whose functions are not fully elucidated. Dysregulation of ADD3 expression or post-translational modification is implicated in diseases including cerebral palsy, hypertension, glioma progression, and other renal and vascular disorders. Gamma-adducin is subject to regulation by phosphorylation and calmodulin binding, affecting spectrin-actin assembly and cytoskeletal dynamics. There are currently no direct drug modulators or therapies targeting ADD3, but its expression and modification status may serve as biomarkers for disease progression, especially in cancer and glomerular disease.
