Overview

Gamma-aminobutyric acid A receptor modulators are compounds that modulate the activity of the GABA<sub>A</sub> receptor, a major inhibitory neurotransmitter receptor in the central nervous system. These modulators can be either positive allosteric modulators (PAMs), which enhance the effect of GABA, or negative allosteric modulators (NAMs), which inhibit it. The GABA<sub>A</sub> receptor is an ionotropic, ligand-gated chloride channel composed of various subunits. Allosteric modulators bind at sites distinct from where GABA binds, altering receptor function without directly activating or blocking it. PAMs are widely used therapeutically for their sedative-hypnotic and anxiolytic properties, while NAMs may have cognitive-enhancing or antidepressant properties in selective cases. Dysfunctional modulation can contribute to neurological diseases such as epilepsy.
