The Gamma-aminobutyric acid receptor subunit alpha2 (GABRA2) is a critical protein component of the heteropentameric GABA-A receptor, which functions as the primary inhibitory neurotransmitter-gated ion channel in the mammalian central nervous system [2, 3]. It forms a ligand-gated chloride channel that, upon activation by GABA, allows the influx of chloride ions into the postsynaptic neuron, leading to hyperpolarization and a subsequent reduction in neuronal excitability [3, 9, 14]. This subunit is highly expressed in brain regions associated with emotional control and reward, such as the hippocampus, amygdala, and striatum, where it plays a pivotal role in mediating anxiolytic and anticonvulsant effects [2, 6, 13]. Genetic variations and mutations in the GABRA2 gene are significantly linked to alcohol and substance use disorders, anxiety, and severe neurological conditions like developmental and epileptic encephalopathy 78 (DEE78) [4, 5, 9, 15]. Pharmacologically, GABRA2 is a key target for benzodiazepines and barbiturates, with current drug development focusing on selective positive allosteric modulators to treat anxiety and epilepsy with reduced sedative and addictive side effects compared to non-selective agents [6, 13, 17, 22].
