Gamma-crystallin D (CRYGD) is a highly stable, monomeric structural protein predominantly found in the vertebrate eye lens. It belongs to the gamma-crystallin family, which is part of the larger ubiquitously expressed crystallin superfamily involved in maintaining lens transparency and optimal refractive properties. Unlike most proteins, gamma-crystallins are synthesized during lens development and then retained without turnover for the organism’s lifetime due to the anucleate state of mature lens fiber cells. Mutations, post-translational modifications (such as deamidation), or age-related changes in CRYGD can induce aggregation, resulting in lens clouding and cataract formation. Though not a classical drug target (such as an enzyme, transporter, or receptor), CRYGD's stability and aggregation propensity are central to cataract pathogenesis, making it of major interest in lens biology and disease research
