Overview

Gamma delta-1 T cell receptor (γδ-1 TCR) is a cell surface heterodimer predominantly expressed by Vδ1 γδ T cells. This receptor recognizes a diverse, often non-peptidic repertoire of antigens independently of classical major histocompatibility complex (MHC) presentation, distinguishing it from the αβ T cell receptor. Upon ligand binding, the γδ-1 TCR associates with CD3 subunits to initiate strong signaling cascades, leading to rapid immune effector responses such as cytotoxicity, cytokine release, and modulation of tissue homeostasis. The Vδ1 subset, characterized by specific chemokine receptor expression (such as CCR2, CXCR1), is enriched in epithelial tissues (skin, gut) and plays dual protective and pathological roles, including antitumor activity, infection response, tissue repair, and, if dysregulated, contribution to inflammatory disorders and fibrosis. The unique ligand recognition flexibility (polyspecificity) of γδ-1 TCR enables responses to a broad array of molecular challenges, making it a promising—though complex—target in immunotherapy for cancer and inflammatory diseases.
