Overview

Gamma-delta (γδ) T-cell receptor ligands are a diverse group of molecules expressed on the surface of cells undergoing stress, transformation, or infection, serving as primary targets for γδ T-cell-mediated surveillance (Vantourout & Hayday, 2013). Unlike conventional αβ T cells, γδ T cells recognize these ligands—including butyrophilins (BTN3A1, BTN2A1), MHC-related proteins (MICA/B), and lipids—in a non-MHC-restricted manner (Karunakaran et al., 2020). This recognition allows for the rapid detection of metabolic changes, such as the accumulation of phosphoantigens in cancer cells or the presence of viral proteins (Rigau et al., 2020). In therapeutic development, these ligands are targeted to redirect γδ T cells to eliminate tumors or infected cells using monoclonal antibodies, bispecific engagers, or small molecules that modulate ligand availability (ImCheck Therapeutics, 2023). Their unique ability to bridge innate and adaptive immunity makes them highly attractive for immunotherapy, particularly in solid tumors where conventional T-cell infiltration may be limited (LAVA Therapeutics, 2023).
