Gamma-delta T-cell receptor (γδ TCR) ligands are a heterogeneous group of molecules expressed by stressed, infected, or neoplastic cells that trigger the activation of γδ T cells, a unique lymphocyte subset bridging innate and adaptive immunity. In humans, the most prominent ligands are phosphoantigens (PAs), such as isopentenyl pyrophosphate (IPP), which accumulate in tumor cells due to dysregulated mevalonate metabolism (Rigau et al., 2020, Science). These PAs are recognized by Vγ9Vδ2 T cells through a mechanism involving the butyrophilin family members BTN3A1 and BTN2A1, which act as molecular sensors of metabolic distress (Karunakaran et al., 2020, Nature). Other ligands include stress-induced proteins like MICA and MICB, which are upregulated on the surface of malignant cells and signal cellular damage (Vantourout & Hayday, 2013, Nature Reviews Immunology). Therapeutic strategies targeting these ligands include small molecules like zoledronic acid that increase PA levels, monoclonal antibodies such as ICT01 that stabilize BTN3A1 complexes, and bispecific engagers that bridge γδ T cells to tumor antigens. Because γδ T-cell recognition is MHC-independent, these targets are highly attractive for developing universal, off-the-shelf cellular and biological therapies with a reduced risk of graft-versus-host disease.
