Overview

The **Gamma-delta T cell receptor Vδ2 chain** is a protein subunit of the gamma-delta (γδ) T cell receptor, which is primarily paired with the Vγ9 chain in human peripheral blood T cells. This forms the Vγ9Vδ2 TCR, the main circulating γδ T cell population in adults. Unlike classical αβ T cell receptors, Vγ9Vδ2 TCRs recognize non-peptide phosphoantigens that accumulate in infected or transformed cells, often presented via butyrophilin 3A1 (BTN3A1). Activation of these cells results in rapid, MHC-independent cytotoxicity against tumor and infected cells, production of cytokines, and crosstalk with other immune effectors, situating them at the intersection of innate and adaptive immunity. Due to these properties, the Vδ2 chain (specifically as part of the Vγ9Vδ2 TCR) is an emerging target in immunotherapy for cancer and infectious diseases[1][2][3][4][5][7].

Mechanism of action

Phosphoantigen mediated activation: Drugs or endogenous metabolites (phosphoantigens) accumulate and are recognized by the Vγ9Vδ2 TCR via presentation through butyrophilin 3A1/BTN3A1, leading to activation of cytotoxic and immune functions[7][1][2]. Direct tumor cell lysis: Upon activation, Vδ2 T cells exhibit direct cytotoxicity against tumor cells[2][5]. Cytokine secretion: Upon stimulation, these cells produce pro-inflammatory cytokines that contribute to immune modulation and anti-tumor response.
