Gamma-glutamyltransferase 7 (GGT7) is a member of the gamma-glutamyltransferase enzyme family, involved in glutathione metabolism and the transpeptidation of amino acids[1][2][3][4]. The enzyme consists of heavy and light chains and interacts with proteins such as CT120, which has roles in lung carcinogenesis[1][2]. GGT7 is normally highly expressed in healthy brain tissue, but expression is reduced in glioblastoma, where lower levels correlate with increased tumor proliferation, anchorage-independent growth, higher reactive oxygen species, and poorer survival outcomes[1]. Increasing GGT7 expression can suppress tumor growth through its antioxidant activity and maintenance of redox equilibrium. GGT7 may serve as a prognostic biomarker and potential therapeutic target in glioblastoma and is implicated in other cancers and metabolic conditions[1][2][6]. Changes in gamma-glutamyltransferase activity can also indicate toxic or preneoplastic states in the liver and kidney[2].
Drugs that upregulate GGT7 may reduce cell proliferation and lower ROS in glioblastoma[1]. Drugs that inhibit, downregulate or genetically silence GGT7 increase cell proliferation in tumor models[1]. Gamma-glutamyltransferase inhibitors in general act by blocking the enzyme's catalytic activities, though none are specifically listed for GGT7.
