Overview

Gamma-parvin (PARVG) is one of three mammalian parvin proteins, alongside alpha-parvin (PARVA) and beta-parvin (PARVB). The parvins are actin-binding proteins found at cell-matrix adhesion sites, particularly in focal adhesions where they interact with components such as integrin-linked kinase (ILK) and paxillin. Gamma-parvin is encoded by the *PARVG* gene and contains two calponin homology (CH) domains, characteristic of actin-binding proteins but structurally distinct from other members of the α-actinin superfamily. Unlike PARVA and PARVB, which are widely studied for their roles in cancer and cell motility, gamma-parvin is primarily noted for its function in promoting leukocyte adhesion and spreading[1][2][3]. Its specific contributions seem more narrowly linked to immune cell adhesion biology, with no established role as a therapeutic target, biomarker, or disease modifier. The functional protein family is evolutionarily conserved, and gamma-parvin shares less sequence identity with alpha- and beta-parvin, indicating distinct roles within cells[1][2][3].
