Gamma-secretase is a multi-subunit, integral membrane protease complex responsible for the intramembrane cleavage of various type I transmembrane proteins. It is best known for its role in processing amyloid precursor protein (APP) to generate amyloid-beta (Aβ) peptides, which are implicated in Alzheimer's disease pathology. The enzyme also cleaves other substrates such as Notch receptors, making it central to multiple signaling pathways and cellular processes. The complex consists of four essential protein subunits: Presenilin (PSEN1 or PSEN2), Nicastrin, APH-1 (Anterior Pharynx Defective 1), and PEN-2 (Presenilin Enhancer 2).
