Overview

Gamma-tubulin complex component 3 (TUBGCP3) is a cytoplasmic protein that is a core constituent of the gamma-tubulin ring complex (γ-TuRC), which mediates microtubule nucleation at the centrosome[1][4]. TUBGCP3 binds directly to gamma-tubulin and is involved in the assembly and function of γ-TuSC and γ-TuRC, which are essential for proper spindle pole formation and cell cycle progression during both mitosis and meiosis[1][3][4]. Loss or depletion of TUBGCP3 disrupts microtubule nucleation, causing mitotic defects, abnormal spindle assembly, cell cycle arrest, and apoptosis[3]. Mutations or dysregulation of TUBGCP3 have been implicated in developmental disorders (including primary autosomal recessive microcephaly), certain cancers, neurodevelopmental defects, and Parkinson’s disease[1][3]. To date, TUBGCP3 itself is not considered a common direct therapeutic target and there are no known interacting drugs or biomarker uses in clinical practice[1].
