Overview

The gamma-tubulin ring complex (γ-TuRC) is a multi-protein complex essential for the nucleation and polymerization of microtubules in eukaryotic cells. It serves as a molecular scaffold that templates the formation of microtubules by anchoring and organizing α/β-tubulin dimers, enabling rapid and spatially controlled polymerization, especially at microtubule-organizing centers such as the centrosome. This process is critical for cell division, cell structure maintenance, intracellular transport, and establishing cell polarity. Dysregulation of microtubule nucleation and polymerization is implicated in cancer (where altered microtubule dynamics facilitate abnormal cell division), neurodegenerative diseases, and nerve injury. γ-TuRC is a validated target for several cytoskeletal drugs used mainly in oncology and neurology, although direct inhibitors are less common than drugs targeting the polymerized microtubule itself[1][3][5][6].
