Overview

Ganglioside GD2 lactone is a conformational variant of the disialoganglioside GD2, featuring an intramolecular ester bond between its sialic acid residues (Source: PMID: 1558593). This modification enhances the immunogenicity of the ganglioside, making it a preferred candidate for the development of conjugate vaccines like GD2-lactone-KLH (Source: J Clin Oncol, DOI: 10.1200/JCO.2001.19.11.2818). GD2 lactone is predominantly expressed on the surface of neuroectoderm-derived tumors such as neuroblastoma, melanoma, and certain sarcomas, while its presence in normal tissues is largely restricted to the central nervous system and peripheral nerves (Source: MSKCC). In oncology, it serves as a critical therapeutic target for monoclonal antibodies and vaccines designed to trigger immune-mediated destruction of malignant cells. Therapeutic strategies targeting this molecule aim to induce antibody-dependent cellular cytotoxicity (ADCC) and complement-mediated lysis (Source: FDA Label for Unituxin). However, targeting this molecule is associated with significant side effects, most notably severe neuropathic pain, caused by the binding of anti-GD2 antibodies to GD2 expressed on sensory nerve fibers (Source: Nature Reviews Cancer, DOI: 10.1038/nrc3694).
