Overview

GAS8 antisense RNA 1 (GAS8-AS1) is a long non-coding RNA (lncRNA) located within the second intron of the GAS8 gene, also referred to as C16orf3[3]. GAS8-AS1 regulates gene expression mainly at the transcriptional and post-transcriptional level, impacting a wide range of biological processes such as cell proliferation, cell cycle progression, migration, invasion, and autophagy[1][3]. Functionally, GAS8-AS1 acts as a tumor suppressor in various cancers, most notably suppressing papillary thyroid carcinoma cell proliferation and tumor growth through interactions with specific microRNAs (such as miR-135b-5p) and downstream effectors (e.g., Cyclin G2)[1]. Beyond oncology, GAS8-AS1 dysregulation has also been implicated in neurodegenerative conditions, with evidence for altered expression in diseases like multiple sclerosis, possibly linking it to immune and inflammatory signaling pathways[2]. GAS8-AS1 is emerging as a promising diagnostic and prognostic biomarker and may represent a novel target for future RNA-based therapies, but it is not a conventional therapeutic target such as a receptor, enzyme, or transporter[3][4]. There are no current drugs directly targeting GAS8-AS1, nor are specific safety concerns identified for it as a therapeutic target.
