Gasdermin D (GSDMD) is a member of the gasdermin family of pore-forming proteins and a key executioner of pyroptosis, an inflammatory form of programmed cell death. Upon activation by inflammatory caspases, the N-terminal fragment of GSDMD forms large pores in the plasma membrane, leading to cell lysis and the release of pro-inflammatory cytokines such as IL-1β and IL-18. GSDMD plays a central role in innate immunity and host defense, but its dysregulation is implicated in various inflammatory diseases and cancers. As such, it is considered an attractive drug target for treating conditions characterized by excessive inflammation.
