Overview

Gastric adenocarcinoma predictive long intergenic non-coding RNA (GAPLINC) is a cytoplasmic long non-coding RNA originally identified in gastric cancer and characterized as an oncogenic lncRNA promoting cell proliferation, migration, and invasion by modulating the expression of CD44 and acting as a molecular sponge for specific microRNAs (including miR-211-3p, miR-661, and miR-135b-5p). GAPLINC is highly upregulated in various cancers—such as gastric, colorectal, renal cell carcinoma, non-small cell lung cancer, glioma, bladder, and osteosarcoma—and its increased expression correlates with poor clinical prognosis and tumor progression. In the immune system, GAPLINC acts as a negative regulator of inflammatory gene expression in macrophages, influencing responses to endotoxin and sepsis by modulating NF-κB localization and activity. Due to its versatile roles in tumorigenesis, metastasis, and immune regulation, GAPLINC is being explored both as a prognostic biomarker and a putative therapeutic target in multiple diseases.
