Gastric cancer associated WDR5 and KAT2A binding long non-coding RNA (GCAWKR)

Target
GCAWKR
Molecular classification
Long non-coding RNA (lncRNA), Molecular scaffold, Chromatin modifier binding partner
01

Overview

Gastric cancer associated WDR5 and KAT2A binding lncRNA (GCAWKR) is a long non-coding RNA (lncRNA) highly upregulated in gastric cancer tissues[1][5]. GCAWKR promotes gastric tumorigenesis by serving as a molecular scaffold for two chromatin-modifying proteins, WDR5 and KAT2A, enhancing their co-localization at target gene promoters and increasing oncogenic histone modifications. Functionally, GCAWKR drives cell proliferation and invasion in multiple gastric cancer models and its expression correlates with poor clinical outcomes, including increased tumor size, metastasis, and advanced disease stage[1][7]. It represents a potential therapeutic biomarker and target for novel anti-cancer strategies focused on disrupting pro-tumorigenic chromatin complexes[1][5][7].

02

Mechanism of action

Not directly applicable as a drug target yet; mechanistically, it promotes carcinogenesis by scaffolding WDR5/KAT2A and facilitating active chromatin marks at oncogenic promoters

03

Biological functions

Acts as a scaffold for WDR5 and KAT2A complexesModulates chromatin modifications on target gene promotersRegulates cell proliferationRegulates cell invasion
04

Disease associations

Cancer (specifically, gastric cancer)
05

Safety considerations

As GCAWKR itself is not yet a clinical drug target, direct safety concerns are not reported; however, targeting lncRNAs may risk off-target effects or disruption of normal epigenetic homeostasis.
06

Biomarkers

High GCAWKR levels are correlated with poor prognosis, larger tumor size, lymph node metastasis, and advanced TNM stage in gastric cancer
