Overview

Gastric mucin proteins, primarily Mucin-5AC (MUC5AC) and Mucin-6 (MUC6), are high-molecular-weight, heavily glycosylated proteins that form the structural backbone of the gastric mucus layer. MUC5AC is predominantly secreted by surface mucous cells, while MUC6 is produced by mucous neck and gland cells, together creating a stratified viscoelastic gel that protects the gastric epithelium from hydrochloric acid, pepsin, and mechanical stress (nih.gov, patsnap.com). These proteins also serve as a critical component of the innate immune system by trapping pathogens such as Helicobacter pylori and maintaining a stable pH gradient through the entrapment of bicarbonate ions (nih.gov, acs.org). In clinical practice, gastric mucins are significant therapeutic targets for cytoprotective agents like rebamipide and misoprostol, which stimulate their production to promote mucosal healing in gastritis and peptic ulcer disease (nih.gov, droracle.ai).
