Overview

The cellular membranes of gastric mucosal cells constitute the primary physical and chemical barrier protecting the stomach wall from autodigestion by gastric acid and pepsin (StatPearls: Physiology, Gastric Barrier, 2023). These membranes are rich in phospholipids and contain specialized proteins such as the H+/K+ ATPase (proton pump) and various G protein-coupled receptors that regulate the secretion of hydrochloric acid and protective bicarbonate (NIH: Gastric Mucosal Protection, 2022). In the context of pharmacology, these membranes are the site of action for cytoprotective drugs like sucralfate, which polymerizes and binds to exudates on the membrane surface to form a protective layer (PubChem: Sucralfate, 2024). Additionally, prostaglandin analogs like misoprostol interact with membrane-bound receptors to enhance mucosal blood flow and mucus production (PubMed: Prostaglandins and the Gastric Mucosa, 2021). Damage to these membranes, often caused by NSAIDs or H. pylori infection, leads to the development of gastritis and peptic ulcers. Therapeutic strategies focusing on this target involve either the direct protection of the membrane surface or the modulation of membrane-bound transporters to reduce the aggressive factors of the gastric environment.
