Gastrin-17 (G17) is a 17-amino acid peptide hormone primarily produced by G cells in the gastric antrum and is the predominant circulating form of gastrin (UniProt: P01350). Its primary physiological role is the stimulation of gastric acid secretion from parietal cells and the maintenance of the gastric mucosa through trophic effects (StatPearls: Gastrin). Glycine-extended gastrin-17 (G17-Gly) is a non-amidated biosynthetic intermediate that was previously considered inactive but is now recognized as a potent growth factor that promotes cell proliferation and inhibits apoptosis in various tissues (PubMed: 10423245). In the context of oncology, both G17 and G17-Gly are implicated in the progression of gastrointestinal and pancreatic cancers, where they act as autocrine or endocrine growth factors (PubMed: 15541573). Therapeutic interventions, such as the G17DT (Poligratide) vaccine, target these peptides by inducing neutralizing antibodies that sequester the hormones, thereby blocking their interaction with receptors like the cholecystokinin B receptor (CCK2R) and inhibiting tumor growth (PubMed: 12105110). This approach is particularly relevant for gastrin-dependent malignancies where traditional chemotherapy may have limited efficacy.
