Target intelligence / Profile preview

Gastrin-17 (often specifically "Little gastrin I" or "Gastrin heptadecapeptide") (Gastrin-17)

Target
Gastrin-17
Molecular classification
Peptide hormone, Regulatory peptide
01

Overview

Gastrin-17 ("little gastrin") is a 17-amino-acid peptide hormone released predominantly by G cells in the gastric antrum in response to food, vagal stimulation, and luminal peptides/amino acids. Its main biological role is to stimulate secretion of gastric acid by parietal cells through activation of the cholecystokinin B receptor, enhance mucosal cell proliferation, and promote GI motility. It is clinically important as an indicator of gastric function and as a biomarker for gastric diseases, especially conditions with altered acid production and certain gastrointestinal cancers. Gastrin-17 is not itself a drug target but is critical for regulating gastric physiology and is often measured in clinical assays.

Mechanism of action

N/A for drugs directly targeting Gastrin-17. Drugs typically modulate secretion or act on its receptor (CCK2R). For receptor antagonists: block CCKB receptor, thereby inhibiting gastrin-mediated acid secretion

04

Disease associations

Cancer (specifically gastrointestinal malignancies)Hypergastrinemia (e.g., in gastrinomas, *Helicobacter pylori* infection, Zollinger-Ellison syndrome)Gastroesophageal reflux disease (GERD)
05

Safety considerations

Hypochlorhydria (risk with elevated gastrin-17 from acid suppression)Tumorigenesis risk with chronic hypergastrinemia

Interacting drugs

None directly for Gastrin-17. However, drugs affecting gastrin secretion include:

Biomarkers

Plasma gastrin-17 levels (used for diagnosis and monitoring of gastric disorders, gastrinomas, monitoring PPI therapy, H. pylori infection)
