The gastrointestinal and respiratory mucosae are specialized tissue layers that line the internal tracts of the body, serving as the primary interface between the internal environment and external agents (StatPearls, 2023). These mucosal surfaces consist of epithelial cells, underlying connective tissue known as the lamina propria, and often a layer of smooth muscle, providing critical barrier, absorptive, and secretory functions (NIH, 2021). In the respiratory tract, the mucosa facilitates gas exchange and traps pathogens via mucus and cilia, while the gastrointestinal mucosa manages nutrient uptake and maintains a complex immunological tolerance to food and microbiota (PubMed, 2022). While not a single molecular target, the mucosa is a major site for drug delivery and the focus of treatments for conditions like inflammatory bowel disease, asthma, and various infections (Nature Reviews Drug Discovery, 2020). Therapeutic interventions often aim to enhance barrier integrity, modulate local immune responses, or provide physical protection to the epithelial surface.
Drugs acting on these tissues typically provide physical protection (cytoprotection), reduce local inflammatory cytokine production, or modulate the activity of mucosa-associated lymphoid tissue (MALT) to restore barrier integrity (StatPearls, 2023).
